摘要 : Ammonia can rapidly accumulate in intensive aquaculture and pose serious threats to fish health. Fish skin functions as a vital immune-related organ and the largest surface barrier against ammonia; however, little is known about the skin’s response to acute ammonia stress. In this study, striped catfish were exposed to the half-lethal ammonia concentration as stress for skin response evaluation. We found the inflammation in fish. Yet, the skin tissue showed subtle signs of damage. Skin tissue ammonia concentration measurement showed ammonia accumulates but lessens in the skin. Further transcriptomic analysis of skin gene function implied that an essential regulation occurs potentially in cell compartments. Transmission electron microscopy–based cellular examination revealed that vacuoles and potential exosomal vesicles can be associated with ammonia regulation during acute ammonia stress. Striped catfish skin epithelial cell line was then used along with an exosome inhibitor to confirm exosomal vesicles and their function in hypothesized ammonia regulation. In conclusion, although acute ammonia stress induces inflammation in striped catfish skin under ammonia stress, active ammonia excretion via exosomes may alleviate ammonia accumulation, thus maintaining skin tissue integrity. This study highlights a new finding of how fish skin responds and protects itself during acute ammonia stress.
