摘要 : Fish skin mucus and its microbiota act as a first line of defense against environmental threats. While environmental changes are known to alter the skin microbiota, the effect of constant mucus shedding and regeneration remains unclear. This study investigates microbiota dynamics following mucus renewal in a controlled laboratory setting. We developed a method to acutely remove skin mucus from one side of the fish and acclimated them in germ-free water for 14 Days. Microbiota communities from mucus-renewal and non-renewal sides were compared using 16S rRNA gene analysis, alongside assessments of skin inflammation and physical activity. Results revealed significant differences in microbiota composition between the two sides, with the renewed mucus showing a dominance of gut-oriented (70%) and potentially pathogenic (20%) genera. Despite this shift, both sides exhibited low inflammatory gene expression and normal physical activity levels. These findings suggest that renewed mucus fails to restore the original microbiota, allowing colonization by gut bacteria, yet without negatively impacting host health. The study highlights the role of continuous environmental exposure in maintaining the natural fish skin microbiota and provides new insights into microbial ecology on fish skin.
